Most ADCs are conjugated through lysine with small molecule drugs, and their measurement is usually based on traditional RPLC/MS platforms, where proteins require DAR determination under denaturing conditions. However, these denaturing conditions can cause the separation of light and heavy chains in cysteine-conjugated ADCs, so cysteine-conjugated ADCs can be analyzed using native mass spectrometry.
