Protein Identification
Protein identification is about determining the correctness of a protein product. Protein molecules are very complex, with many characteristic parameters. The most commonly measured parameters currently include the molecular weight of the protein, the isoelectric point of the protein, protein structure, biological function of the protein, and immunological reactions.
The molecular structure of proteins is divided into four levels: Primary structure refers to the sequence of amino acids in the protein polypeptide chain; Secondary structure refers to highly ordered local substructures on the actual peptide backbone, such as α-helix and β-sheet; Tertiary structure refers to the three-dimensional structure formed by the spatial arrangement of multiple secondary structures; Quaternary structure refers to the three-dimensional structure formed by the aggregation of two or more individual polypeptide chains (subunits). In protein identification, commonly used parameters related to protein structure include primary/secondary structure, crystal structure, peptide profile, and N/C termini.
