Therefore, MALDI-TOF MS becomes an ideal tool for mass peptide mapping analysis. When you need to verify the theoretical sequence of protein (peptide) samples using MALDI TOF, we will first enzymatically digest the samples obtained, then confirm the peptide fingerprint map with MALDI MS, validate the peak map with MS/MS, and finally compare it with the theoretical sequence you provided to obtain the peptide coverage map. Advantages and Disadvantages of MALDI TOF for Mass Peptide Mapping Analysis:
Advantages:It is a classic method for protein identification, with a simple algorithm and fast speed. It was widely used before the advent of tandem mass spectrometry identification.
Disadvantages:Peptides with similar mass increase the difficulty of matching, and it is impossible to identify mixed proteins. It is not suitable for species with incomplete databases for protein identification. It cannot analyze post-translational modification sites and is not easy to connect with liquid chromatography.
