Amino Acid Composition Analysis

Before delving into the analysis of amino acid composition, let's briefly introduce a few definitions. Amino acids are organic compounds containing amine (-NH2) and carboxyl (-COOH) functional groups, along with a side chain (R group) specific to each amino acid. A peptide (from Greek πεπτός, 'digested' peptós; derived from πέσσειν, 'to digest' péssein) is a short chain of two to fifty amino acids linked by peptide bonds. A polypeptide is a longer, continuous, unbranched peptide chain that can contain up to about fifty amino acids. Polypeptides/peptide chains containing more than fifty amino acids are referred to as proteins.

Proteins are composed of one or more polypeptides arranged in a biologically functional manner, and they can usually bind to ligands (such as coenzymes and cofactors), another protein, other macromolecules (such as DNA or RNA), or more complex macromolecular assemblies. Antibodies (Ab), also known as immunoglobulins (Ig), are large Y-shaped proteins mainly produced by plasma cells and used by the immune system to neutralize pathogens like pathogenic bacteria and viruses.

Amino Acid Composition Analysis

Amino acid composition analysis is the gold standard for accurately quantifying samples such as polypeptides, proteins, antibodies, etc. Typically, we aim to know the amino acid composition of a sample before determining its sequence information, as this helps identify errors in the sequencing process and allows for corrections. Understanding the amino acid composition of a sample can also assist in determining which protease is suitable for hydrolyzing the sample and in identifying the incorporation of low-level non-standard amino acids (such as norleucine) in proteins. The common method for determining amino acid composition is often referred to as Amino Acid Analysis. It can be used to determine the amino acid composition, components, and content of amino acids, peptides, polypeptides, proteins, and antibody samples. Amino acid analysis is also known as amino acid composition analysis. It allows for both qualitative and quantitative determination of the amino acid composition in your samples.

About the Sample

Sample Purity
Biotech BioTech can analyze most types of samples, including amino acids, peptides, polypeptides, proteins, antibodies, etc.;
Please avoid high salt concentrations in the sample;
Do not use buffers containing large amounts of primary amines, such as TRIS or glycine, as they may interfere with the ninhydrin reaction, leading to inaccurate quantification of specific amino acids;
Avoid fingerprints and dust in the sample, as these substances contain large amounts of human proteins, which may affect the test results.

Sample Amount
The sample amount should be >2 mg whenever possible.
Provide a sufficient amount of solid material to accurately weigh the sample during our experiments.
*You can choose one or three amino acid determination replicates. When you need very accurate protein concentration determination, we recommend three biological replicates.
