Glycosylation, especially N-glycosylation, is a common post-translational modification that regulates the localization, function, activity, lifespan, and diversity of proteins in organisms. Research on N-glycosylation sites is one of the important prerequisites for understanding the function of glycan chains. In the information related to N-glycosylation, the sites where N-glycosylation occurs are the basis for understanding its glycan function. Besides traditional biochemical methods such as site-directed mutagenesis for studying glycosylation sites, mass spectrometry also plays an important role in the identification of N-glycosylation sites. MALDI TOF MS and nano LC-ESI-MS/MS are commonly used for the identification of N-/O-glycosylation sites in antibodies. After receiving samples, we first perform enzymatic digestion on the samples to release the Fc domain and Fab of the antibodies, then separate the glycosylated fragments and confirm the glycosylation sites via MALDI TOF MS or nano LC-ESI-MS/MS.

Glycosylation site identification process
(1) Perform enzymatic digestion on the sample using trypsin or other appropriate proteases;
(2) Select suitable chromatography columns to separate N-glycopeptides, O-glycopeptides, and other peptides;
(3) Perform deglycosylation treatment;
(4) Analyze peptide maps via MALDI-TOF MS, ESI-MS, or MS/MS to identify theoretical glycosylation sites;
(5) Determine each N-/O-glycan site from the peptide maps;
(6) Compare glycosylation sites across different batches of samples.
