Matrix-assisted laser desorption/ionization (MALDI) is a soft ionization technique used in mass spectrometry for the identification of protein samples that may produce fragments through ionization methods. Compared to another soft ionization technique, electrospray ionization (ESI), MALDI generates fewer multiply charged ions and allows for full-spectrum analysis of protein samples without the need for additional deconvolution steps. However, for proteins with larger molecular weights, the difference between average mass and accurate mass is significant. Therefore, for proteins with molecular weights greater than 25 kDa, high-resolution mass spectrometry identification strategies are recommended.
