Currently, two main types of mass spectrometry are widely used for protein identification: one is MALDI-TOF, which directly measures molecular weight. The ions produced by the MALDI source are mostly singly charged, and the peaks in the mass spectrum correspond directly to the mass of each component in the sample, allowing the calculation of molecular weight from the spectrum without deconvolution. The other is ESI-MS high-resolution mass spectrometry, which analyzes multi-charged signals obtained through electrospray and then deconvolutes the signals to obtain precise molecular weight values. Both methods can be used for measuring the molecular weight of protein complexes and protein-small molecule complexes.
