Targeted Proteomics
Targeted proteomics is a branch of science that has evolved from proteomics, focusing on the quantitative study of specific target proteins. Compared to untargeted proteomics, targeted proteomics selectively acquires mass spectrometry data for specific ions, making it suitable for analyzing large numbers of samples with greater accuracy, sensitivity, and reproducibility. Targeted proteomics can be applied to system-level studies of post-translational modifications, protein conformations, protein-protein interactions, dynamics, as well as metabolic and signal transduction pathways.
Targeted proteomics quantitative techniques primarily include MRM (multiple reaction monitoring) and PRM (parallel reaction monitoring). MRM, also known as SRM (selected reaction monitoring), uses triple quadrupole mass spectrometry (QQQ-MS) to collect signals that meet the target ion criteria, thereby achieving relative or absolute quantification of target proteins. PRM is derived from MRM, combining the high selectivity of quadrupoles with the high resolution and accuracy of Orbitrap. It measures all fragment ions after fragmenting selected parent ions, offering better detection sensitivity and interference resistance.
