DIA Quantitative Proteomics

DIA (data-independent acquisition) refers to a data-independent scanning mode, which is a holographic mass spectrometry data acquisition mode based on the Orbitrap electrostatic field. After the first-level mass spectrometry detection, DIA fragments all parent ions within a specific mass-to-charge ratio range, collects fragment ions of all parent ions, and quickly scans all fragment ions within adjacent wide parent ion windows, thereby performing qualitative and quantitative analysis of proteins. Compared to DDA (data-dependent acquisition mode), DIA offers better accuracy and reproducibility. SWATH technology is a type of DIA technology that combines the high-throughput detection characteristics of shotgun proteomics with the advantages of PRM precise quantitative analysis, enabling quantitative measurement of almost all detectable molecules in complex samples.
