Sphingosine-1-phosphate (S1P) is a signaling sphingolipid, a product of sphingosine kinase phosphorylating sphingosine. S1P is dephosphorylated back to sphingosine by sphingosine phosphatase and irreversibly degraded by sphingosine phosphate lyase. Many cells can secrete S1P, which then influences other cells in an autocrine or paracrine manner.
Sphingosine-1-phosphate has a wide range of biological effects. It has been shown to enhance cell growth and inhibit normal apoptosis responses. Numerous studies indicate that S1P promotes tumor growth, anti-apoptosis, tumor angiogenesis, and metastasis, as well as enhances angiogenesis. By promoting the release of calcium from internal stores, S1P increases intracellular calcium levels. Moreover, S1P regulates pro-inflammatory cytokines, Toll-like receptor signaling, and the production of inflammation, which are associated with diabetes, obesity, and metabolic syndrome.
