Non-targeted Metabolomics

Metabolic networks are incredibly complex, with human metabolic networks being particularly intricate. Current research on signaling pathways is mostly focused on a very small area of the metabolic network. Genomics and proteomics have uncovered some regulatory pathways, but metabolites are directly related to the metabolic network. Choosing research targets from the vast array of metabolites is like finding a needle in a haystack. Untargeted metabolomics, also known as discovery metabolomics, primarily involves comparing the metabolomes (all metabolites within a biological entity) of control and experimental groups to identify differences. Metabolomics research can help researchers select metabolites from numerous candidates, providing an 'aerial view' to clearly identify differential metabolites. By reverse-engineering known metabolic pathways, researchers can identify regulatory enzymes and genes, aiding research into disease mechanisms and effective treatment mechanisms.

Untargeted Metabolomics Analysis Techniques

1. GC-MS (Gas Chromatography-Mass Spectrometry) is a classic technique in metabolomics research, known for its mature and stable technology, high resolution, and relatively complete database, making qualitative analysis more accurate. Its drawbacks mainly include complex sample processing and challenges in qualitative and quantitative analysis of non-derivatizable substances, limiting its broader application.
2. LC-MS (Liquid Chromatography-Mass Spectrometry) has advantages in simple sample preparation and pre-treatment, good experimental reproducibility, high resolution, and a wide range of separation and analysis. Its drawbacks mainly lie in an incomplete database, making qualitative analysis relatively difficult.

About Samples

1. Microbial and cell samples: Rapidly quench metabolic activity while maintaining cell integrity
2. Animal body fluids (such as urine, blood, tissues, organs, saliva): Quickly pre-treat after sampling, such as adding anticoagulants, preservatives, and immediately freeze at -80°C
3. Plant samples: Quickly freeze with liquid nitrogen after collection, then store at -80°C, 200mg/sample
4. Serum samples: 500μl/sample (not less than 200μl/sample), avoid repeated freeze-thaw cycles. (Allow blood to clot for 30 minutes in a centrifuge tube. Centrifuge to collect the supernatant into a clean centrifuge tube, centrifuge for 5 minutes at 8000rpm, aliquot supernatant into cryovials, and ship frozen at -80°C.)
5. Urine samples: 1ml/sample, preferably more (Directly aliquot urine into centrifuge tubes, 1ml per tube, add one drop (about 10μl) of 1/100 (w/v) azide sodium, freeze at -80°C and ship)
