LC-MS High-Resolution Mass Spectrometry Molecular Weight Detection
Liquid Chromatography-Mass Spectrometry (LC-MS) is apowerful technique combining Liquid Chromatography (LC) and Mass Spectrometry (MS)for analysis,allowing the separation,detection, andquantification of varioustypes ofcompounds in mixtures.High-resolution mass spectrometry in LC-MS is amass spectrometry technique with higheraccuracy in measuring molecular weights,making it particularly suitable forthe detection of complexandhighly similar molecular weightmacromolecular compounds.
High-resolution mass spectrometry molecular weightdetection in LC-MS plays a significant role in biomedical research, drug discoveryand development, contributing importantly toresearch. Molecular weight is an essential quality control metric,allowing accurate, rapid, and sensitive measurement of molecular mass,which can largely determine the authenticity and issues of bioproducts..BTP (Biotechnology)has establishedCNAS/ISO9001 dual qualitysystemcertificationsystem;,and based on the principles of molecular weight determination (mass spectrometry), has developed molecular weight identification services for different categories of bioproducts, enabling high-resolution molecular weight measurement of proteins, antibodies, vaccines, peptides, recombinant collagen, and other bioproducts.High-resolution mass spectrometry molecular weightdetection in LC-MSfollows the basic process:

Experimental Instruments
- High-performance liquid chromatograph: Ultimate 3000 (Thermo Fisher Scientific)
- Electrospray-Quadrupole Time-of-Flight Mass Spectrometer: AB SCIEX TripleTOF 5600 (AB SCIEX)
Technical Advantages
- High-resolution mass spectrometry can accurately measure the molecular weight of protein samples (10-250 kDa), with a precision of 1 Da;
- It can analyze the modification states of proteins (such as phosphorylation, small molecule drug binding, etc.), and perform relative quantitative analysis of these modifications;
- It can achieve highly sensitive, high-resolution, and high-accuracy detection of the native, reduced, and deglycosylated molecular weights of antibodies, as well as detection of antibody-drug conjugates.
Case Illustration
Under the influence of an electric field, the sample solution forms highly charged mist-like droplets, which undergo ionization as they move toward the mass analyzer, resulting in singly or multiply charged ions. The different charged ions obtained are deconvoluted to yield the final protein molecular weight.
The figure shows the mass spectrum of different charged ions of the sample and the deconvoluted molecular weight, with the x-axis representing molecular weight and the y-axis representing signal intensity.
