DIA-PRM Proteomics
DIA (data-independent acquisition) refers to a data-independent scanning mode, which is a holographic mass spectrometry data acquisition mode based on the Orbitrap. Compared to DDA (data-dependent acquisition), DIA technology divides the entire mass spectrometry scan range into several windows, selects, fragments, and detects all ions in each window, offering better accuracy and reproducibility. PRM (parallel reaction monitoring) is an ion monitoring technique based on high-resolution and high-precision mass spectrometry. It is currently the mainstream method for targeted proteomics data acquisition, achieving targeted relative or absolute quantification of target proteins/modification peptides by selectively detecting specific peptides or target peptides (such as post-translational modification peptides). Combining DIA and PRM for proteomics research enables detection of large sample sizes or complex samples and precise quantification of target proteins, especially improving the detection rate of low-abundance target proteins.
