PCT-DIA Proteomics
PCT (Pressure Cycling Technology) is an efficient biological sample preparation technology developed by the American company PBI. It uses alternating cycles between atmospheric and ultra-high (liquid) pressure (35,000 PSI or higher) to repeatedly break cells, enabling the extraction of important biomolecules like proteins from various samples, including human, animal, plant, and microbial cells and tissues. DIA (data-independent acquisition) is a holistic mass spectrometry data acquisition mode based on the electrostatic field Orbitrap. Compared to DDA (data-dependent acquisition), DIA technology divides the entire mass spectrometry scan range into several windows, selecting, fragmenting, and detecting all ions within each window, offering better accuracy and reproducibility. The combination of PCT and DIA technologies enables proteomic research on paraffin-embedded or formalin-fixed tissue samples, as well as trace samples.
