Prokaryotic Transcriptome Sequencing

The transcriptome is the complete set of mRNA within a cell at a specific developmental stage or physiological condition. Transcriptome sequencing is widely used as a technical means to study the transcriptome in biological science research. Analyzing the transcriptome information of biological samples helps to study the gene transcription and transcriptional regulation in cells. In prokaryotes, the vast majority of RNA is rRNA, with mRNA accounting for only 1% to 5% of the total RNA. Therefore, to sequence the transcriptome of prokaryotes, mRNA must be purified. Moreover, prokaryotic mRNA does not have a poly A tail like eukaryotic mRNA, so it cannot be directly purified using oligo T. Generally, during prokaryotic transcriptome sequencing, a method to remove rRNA is used to construct the library.
