Oligonucleotide Purity Analysis

Oligonucleotides, including deoxyribonucleic acid (DNA) and ribonucleic acid (RNA), have broad applications in various fields such as gene therapy, biological probes, and diagnostic testing. For example, oligonucleotides canDNAbe introduced into immune cells to genetically modify them to express chimeric antigen receptor proteins for cell-based immunotherapy. Oligonucleotides synthesized using solid-phase synthesis and other techniques may introduce trace impurities at various steps of the synthesis cycle. Therefore, analyzing and testing the purity of oligonucleotides is particularly important to ensure efficacy, accuracy, safety, and bioactivity during the production and research process.

Ion-Pair Reverse-Phase High-Performance Liquid Chromatography (IP RP HPLC) for Oligonucleotide Purity Analysis

 

SDS-PAGE is a traditional purification method that can be used to separate oligonucleotides based on size. However, if the oligonucleotides are of similar size and certain modifications are even inseparable, higher-resolution high-performance liquid chromatography is required. The most popular oligonucleotide purification technique is Ion-Pair Reverse-Phase High-Performance Liquid Chromatography (IP RP HPLC). A key advantage of IP RP HPLC is that it can be directly coupled with mass spectrometry for detailed mass characterization of oligonucleotides. BioTech Pack (BTP) has developed and validated an oligonucleotide purity analysis method based on high-resolution liquid chromatography platforms. This method can select appropriate column lengths, mobile phase flow rates, separation temperatures, and ion-pair reagent buffer compositions according to different oligonucleotide samples to identify impurities such as n-1 short chains, n+1 long polymers, incomplete sulfation, and phosphodiester by-products caused by failed reactions.
