Oligonucleotide Molecular Weight Analysis

Oligonucleotides are molecules composed of relatively short nucleotide units, typically consisting of 2 to 20 nucleotide units. Due to their unique properties, they are increasingly used in disease diagnosis and treatment. They can introduce DNA into immune cells, genetically modify the cells to express chimeric antigen receptor proteins, and achieve cell-based immunotherapy. The molecular weight of oligonucleotides is a very important attribute, and oligonucleotide molecular weight analysis can be used to determine the type and purity of oligonucleotides, evaluate the quality of oligonucleotides, and optimize synthesis methods. In addition, it can be used for the design of drug delivery systems and gene therapy research, which is of great significance for biological research and drug development.

Oligonucleotide Molecular Weight Analysis

Matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI TOF) is an important tool for oligonucleotide molecular weight analysis. MALDI TOF uses a laser with a chemical matrix to ionize oligonucleotide samples, then accelerates the ions through a flight tube to a detector, which measures particle counts as a function of time. The flight time is proportional to the mass of the molecule. MALDI TOF has high throughput, but due to its ionization efficiency and separation resolution, there is a risk that photosensitive modified oligonucleotides may be damaged by the strong laser source. Therefore, it is suitable for analyzing oligonucleotides with fewer than 50 bases.

Another classic method for oligonucleotide molecular weight analysis is electrospray ionization mass spectrometry (ESI-MS). This technique applies high voltage to generate aerosols from liquid samples and ionizes target oligonucleotide molecules into multiple charge states represented by different mass spectra, which can be deconvoluted into parent peaks to identify oligonucleotides and potential impurities. ESI-MS is an ideal tool for analyzing oligonucleotides larger than 50 bases and is also suitable for analyzing photosensitive oligonucleotides due to its milder ionization conditions.
