2D-DIGE Quantitative Proteomics
2D-DIGE (Two-dimensional fluorescence difference gel electrophoresis) is a new quantitative proteomics technique developed from traditional two-dimensional electrophoresis. The principle of separating mixed proteins in 2D-DIGE is consistent with two-dimensional electrophoresis, utilizing differences in the isoelectric point and molecular weight to separate protein mixtures. At the same time, applying the sensitivity of fluorescent dyes and internal standards, it significantly outperforms traditional two-dimensional electrophoresis in quantitative proteomics research. The fluorescent dyes used in DIGE are Cy2, Cy3, and Cy5, which can react with the amino group of the lysine side chain of proteins to label them. The isoelectric point and molecular weight of labeled proteins remain unaffected. After mixing labeled proteins in equal amounts for two-dimensional electrophoresis, differences between gels can be eliminated through Cy2 internal standard matching, while changes in protein expression levels are reflected by the intensity of different fluorescence from Cy3 and Cy5. As a classic method in quantitative proteomics, 2D-DIGE is widely applicable and suitable for various samples.
