Case Illustration
We use a direct analysis method of O-glycopeptides for O-glycosylation site analysis. First, according to the theoretical amino acid sequence information of the protein, an appropriate protease is selected to cleave the protein into peptides. Then, raw mass spectrometry data is collected by liquid chromatography-high resolution mass spectrometry. This data is imported into the O-glycan database, and O-glycan modification is added as a variable modification in the data analysis. Finally, the sites within the protein where O-glycosylation modifications have occurred are identified.
