Glycan Analysis

For the relative quantification detection of glycoforms in glycosylated proteins, Biotech Co., Ltd. provides glycoform relative content analysis services based on MALDI-TOF/LC-MS. Due to the diversity of O-glycosylation modifications and the absence of a universal enzyme to cleave all O-glycans, our glycoform analysis currently targets only N-glycosylated proteins.

The strategy for glycoform analysis services is as follows: Use PNGase F enzyme to completely digest N-glycans from the target protein, label the N-glycans with 2-AA, and specifically enrich N-glycans using SPE columns. Depending on the complexity of glycosylation of the target protein, select MALDI-TOF-TOF or LC-MS/MS to detect 2-AA labeled N-glycans, identify N-glycans using GlycoWorkbench software (CFG database), and analyze the relative content of this glycoform based on the mass spectrum peak area of the respective glycan.

About Samples

Protein Glycoform Analysis Sample Requirements

1. If you are providing tissue samples, please send them to us on dry ice.
2. If you are providing protein samples, use a standard tissue or cell lysis buffer for protein extraction.
3. Sample transportation: Please use an adequate amount of dry ice for transportation and opt for a faster postal service to minimize the possibility of sample degradation during transit.
