Shotgun Proteomics
Shotgun proteomics refers to the use of bottom-up proteomics techniques to study all proteins in complex mixtures such as serum, urine, and cell lysates. It combines high-performance liquid chromatography (HPLC) technology and mass spectrometry (MS). The most unique aspect of shotgun proteomics is that it allows for the identification and quantification of multiple proteins with minimal protein separation.
The purpose of shotgun proteome analysis is to identify as many proteins as possible in a sample. Shotgun proteome analysis utilizes shotgun technology for comprehensive protein analysis, where the basic method involves enzymatic digestion of proteins in a mixture, followed by separation of peptides through liquid chromatography (LC), and then identification of peptides using tandem mass spectrometry (MS/MS) to achieve protein identification. Shotgun proteome analysis allows for the identification of all proteins in a sample, capable of simultaneously identifying hundreds to thousands of proteins.
