Non-targeted Metabolomics Detection of Gas and Liquid Phase

Non-targeted metabolomics, as opposed to targeted metabolomics, aims to study both known and unknown metabolites, identifying all detectable metabolites to analyze overall metabolic characteristics and discover the metabolic mechanisms behind phenomena. Non-targeted metabolomics is a powerful tool for large-scale early diagnosis and the discovery of new biomarkers. Through the use of techniques capable of detecting a large number of metabolites, non-targeted metabolomics analysis can quickly provide insights into the metabolic characteristics of samples.

Gas chromatography-mass spectrometry (GC-MS) and liquid chromatography-mass spectrometry (LC-MS) are both powerful metabolomics analysis techniques. GC-MS is known for its high sensitivity, peak resolution, and reproducibility, and it is widely used in the metabolic analysis of various biological fluids such as urine and blood. The availability of GC-MS electron impact (EI) spectral libraries further facilitates the identification of diagnostic biomarkers and aids in elucidating the mechanisms of biological or pathological variations. Coupled with time-of-flight mass spectrometry (TOF-MS), the novel comprehensive two-dimensional GC (GC×GC) can detect over 1200 compounds in a single analytical run.

LC-MS is the primary method for detecting drugs and their major metabolites in vivo, providing sufficient sensitivity, specificity, and molecular structure information for the qualitative determination of drugs and metabolites. Compared to GC-MS, LC-MS is more widely applicable in analyzing thermally unstable, non-volatile, polar, ionic, and large compounds. LC-MS has become the preferred tool for measuring small molecule components of cell metabolism, capable of simultaneously analyzing dozens to hundreds of chemicals.
