Protein Identification Process
Based on the Shotgun method: Protein bands or protein solutions are enzymatically digested into peptide mixtures, which are then separated by high-performance liquid chromatography into several simpler components. These are analyzed in a high-resolution mass spectrometer. The search software compares the secondary mass spectrometry information with the corresponding database, combines matching scores and mismatch filtering, and determines the exact sequence of the peptides, thus assembling the complete sequence of each protein to achieve protein identification.
Pull-down Target Protein Identification Process: Express and purify bait proteins labeled with GST, polyHis, or Biotin, use immobilized affinity ligands to stably bind bait proteins to solid-phase matrices, then incubate the bait proteins with the cell lysate to be tested. During this process, bait proteins interact and bind with target proteins. By washing away non-specifically bound contaminant proteins, target proteins interacting with bait proteins can be obtained. Then, use mass spectrometry analysis to identify the target proteins.
