iTRAQ Quantitative Proteomics and TMT Proteomics
Labeled quantitative proteomics refers to the study of labeling and quantifying all proteins across multiple different samples. iTRAQ quantitative proteomics and TMT proteomics utilize various isotopic labels to achieve qualitative and quantitative analysis of proteomes from multiple samples.
iTRAQ and TMT are peptide in vitro labeling quantitative techniques developed by Thermo Company and AB SCIEX Company, respectively. They are currently the two most widely used labeling quantitative proteomics techniques.
iTRAQ technology (Isobaric Tags for Relative and Absolute Quantification) is a proteomics quantitative research technology based on in vitro isotope labeling. It is currently mainly used for relative quantitative analysis of proteins among different samples. iTRAQ quantitative proteomics can combine iTRAQ technology with tandem mass spectrometry technology to identify differential proteins in different proteomes in a high-throughput manner and accurately perform relative and absolute quantification. iTRAQ technology uses 4-8 types of isotopic tags to conduct quantitative analysis of proteins in 4-8 samples.
TMT quantitative proteomics and iTRAQ quantitative proteomics have slight differences in tag molecular structure, but they have similar principles. Both can be used to label all peptides from different samples, using different isotopic reagents to label peptides generated after protein digestion, and react with amino (N-terminal and lysine side chain groups) to achieve linkage. They can be applied for quantitative analysis at the whole proteome level across multiple different samples. The difference is that TMT can label up to 16 different biological samples simultaneously, while iTRAQ can label up to 8 different biological samples simultaneously.
