Host Cell Protein Assay

Host Cell Protein Assay (HCP Assay) is a technique used to evaluate the levels of residual exogenous proteins from the host cells used in the production of biologics. These host cell proteins (HCPs) may be introduced during the manufacturing process of biopharmaceuticals, and despite purification steps, may still remain in the final product. The presence of HCPs can cause a range of issues such as affecting the safety and efficacy of the drug, potentially triggering immune responses or interfering with the intended function of the biologic. Therefore, HCP assay plays a crucial role in the quality control of biologics. In biopharmaceutical processes, cell factories such as bacteria, yeast, insect, or mammalian cells are commonly used to produce recombinant proteins. These cells synthesize not only the target protein but also express other 'background' proteins. When the drug product is purified, although most of the background proteins are removed, some host cell proteins may still remain. These residual host cell proteins can adversely affect patients, such as causing overreaction of the immune system or altering the pharmacokinetic properties of the drug. Therefore, scientists are committed to developing precise and sensitive methods to assay host cell proteins to detect and quantify these trace proteins.

 

A major application area for host cell protein assay is the quality control of biologics. During the production of these biologics, HCP assay can ensure the safety and efficacy of the drug. Additionally, in new drug development, this assay can help optimize manufacturing processes, monitor the effectiveness of different purification steps, and confirm the purity and quality of the final product. Regulatory agencies such as the FDA and EMA have strict requirements for the detection and control of host cell proteins, so HCP assay is also a part of pharmaceutical companies' compliance.

 

There are various technical methods for host cell protein assay, among which ELISA (Enzyme-Linked Immunosorbent Assay) is one of the most commonly used methods. ELISA has high sensitivity and specificity and can detect multiple host cell proteins. However, developing specific ELISA kits for each biologic can be time-consuming and costly. Mass spectrometry has gained attention in recent years for HCP assay. Mass spectrometry offers high resolution and a wide dynamic range, providing more detailed protein identification information.

 

In mass spectrometry, common strategies for host cell protein assay include Data Dependent Acquisition (DDA) and Data Independent Acquisition (DIA). DDA methods achieve protein identification and quantification through selective fragmentation of ions, while DIA methods obtain fragmentation spectra of all peptide segments simultaneously, improving the comprehensiveness and accuracy of detection. Mass spectrometry not only helps identify and quantify host cell proteins but also provides information for developing new purification processes.
