How to calculate the protein loading amount when performing SDS-PAGE?

The goal of determining the sample loading amount is to ensure that the protein bands after electrophoresis are clearly visible, but not so concentrated that the bands become fuzzy.

First, you need to know the concentration of protein in your sample. This is usually achieved through protein concentration determination methods such as Bradford, BCA, or Lowry assays. These methods provide a measurement of protein concentration, typically in micrograms per microliter (µg/µl). In general, for most SDS-PAGE experiments, the sample loading amount per well is typically between 10-50 micrograms. The specific loading amount depends on your experimental objectives and the nature of your sample.

Once you have determined the protein concentration in your sample and the desired loading amount, you can calculate the required loading volume.

The formula is: Loading volume (µl) = Loading amount (µg) / Protein concentration (µg/µl).

In SDS-PAGE experiments, the sample needs to be mixed with loading buffer and then boiled at high temperature to denature the proteins. Therefore, you need to consider the volume of the loading buffer to ensure that the volume of your sample plus the loading buffer does not exceed the capacity of the gel well.

Below is a specific example to illustrate:

1. Suppose your protein concentration is 2 µg/µl, and you want to load 20 µg, then the volume you need to load is 20 µg / 2 µg/µl = 10 µl.

2. If the volume of your loading buffer is 5 µl, then your total loading volume is 10 µl + 5 µl = 15 µl, which should fit in your gel well.
