Mammalian Non-targeted Lipidomics

As major components of cell membranes and energy sources, lipids play an important role in both cellular and physiological energy homeostasis. The main biological functions of lipids include storing energy, signal transduction, and serving as structural components of cell membranes. The composition of lipids varies among different tissues. In animal tissues, the composition of structural lipid components (such as phospholipids) is relatively constant under normal physiological conditions. In contrast, the proportion of simple lipids, especially triacylglycerols, can change significantly according to the animal's physiological and nutritional status. The composition of plasma lipids is particularly important as they supply fatty acids to all tissues. The proportion of phosphatidylserine in brain phospholipids is higher than in other tissues. In the liver, PC and PE are very abundant.

Since lipid metabolism is regulated by a network, any changes in synthesis or degradation pathways can lead to alterations in the entire lipid homeostasis of the network. Untargeted lipidomics analysis provides a basis for studying the mechanisms of the entire system following specific stimuli. Moreover, in certain diseases or pathological conditions, identifying altered types and quantities of lipids can reveal the mechanisms of that disease. Additionally, lipidomics can be used for the discovery of biomarkers for various diseases, including diabetes and obesity, cardiovascular diseases, non-alcoholic fatty liver disease, psychiatric disorders, and cancer.

Instruments used for the identification and quantification of mammalian lipidomics include MS, GC, HPLC, or LC-MS, GC-MS. The combination of chromatographic methods (such as GC or HPLC) with MS can greatly increase lipid coverage. LC-MS is one of the widely used techniques in mammalian lipidomics and can be used for qualitative and quantitative analysis of various lipids in mammals, including CER, ChE, CL, DG, FA, LPC, LPE, MG, PA, PC, PE, PG, PS, SM, TG, etc.
