Mass spectrometry imaging (MSI) or imaging mass spectrometry (IMS) is a promising analytical technique that utilizes mass spectrometry to determine molecular mass and visualize the spatial distribution of specific chemical components such as biomarkers, metabolites, peptides/proteins. As a powerful analytical method, matrix-assisted laser desorption/ionization (MALDI) offers spatial resolution and molecular specificity, aiding in label-free tracking of endogenous and exogenous compounds. Similar to immunohistochemistry, MSI can provide high-throughput scanning of compounds without the need for antibodies.

In MALDI-MSI mass spectrometry imaging, the target tissue is sliced and embedded in an organic matrix during sample preparation. The matrix facilitates the desorption and ionization of target compounds under the action of a UV laser beam. The mass-to-charge ratio of the resulting ions is measured by the mass spectrometer in a programmed array at the ablation point. Multiple analytes can be identified and quantified simultaneously. Therefore, MALDI-MSI can provide a rapid snapshot of the molecular distribution at a specific location on the tissue surface.
