Glycosylation Sequencing
Glycan sequencing is a technique used to analyze the structure of glycans (sugar chains) in biomolecules. It is mainly used to study the glycan composition and structure of glycoproteins and glycans.
1. The main steps of glycan sequencing include:
1. Sample Preparation:
Target glycan molecules are extracted from biological samples, often requiring multiple chemical processing steps to remove interfering substances.
2. High-Performance Liquid Chromatography (HPLC):
Different glycans are separated using HPLC for individual analysis.
3. Mass Spectrometry Analysis:
Mass spectrometry techniques, such as tandem mass spectrometry (MS/MS), are used to determine the mass and structure of glycans. This helps identify glycan composition, branching patterns, and linkage types.
4. Data Analysis:
By analyzing mass spectrometry data, different glycan structures in the sample can be identified, and a structural map of glycans can be established.
