How to perform liquid chromatography analysis?

1. Sample Preparation:

3. Mobile Phase Selection:

The mobile phase is usually water, organic solvents, or their mixtures, and may include additives such as buffers, pH adjusters, and ion pairs to optimize separation.


4. Instrument Configuration:

A liquid chromatograph includes an injector, a pump, a chromatographic column, a detector, and a data processing system. Depending on experimental needs, appropriate instruments such as high-pressure constant flow pumps, gradient elution devices, UV-Vis detectors, and fluorescence detectors should be configured.


5. Experimental Operation:

The sample solution is injected into the chromatographic column at an appropriate rate, where the sample components are separated between the stationary phase and the mobile phase. The detector detects the eluted compounds, and the computer system processes the data to obtain quantitative parameters such as peak area or peak height.


6. Data Processing:

In the final step of liquid chromatography analysis, the obtained data needs to be processed, such as quantitative analysis, peak identification, and chromatogram plotting. Typically, liquid chromatography data processing software like Chromeleon, Empower, and Origin can be used.


Liquid chromatography analysis requires the cooperation of instruments and also demands a high level of experimental skills and operational proficiency from the experimenter. Strict control of parameters such as flow rate, temperature, and elution time is necessary to obtain accurate analysis results.
