What to do if the sample contains salt before mass spectrometry?

The presence of salt in samples may interfere with mass spectrometry analysis, affecting the accuracy of the results. If the sample contains salt, the following methods can be used for treatment:


1. Desalting Treatment:

Use desalting columns (such as PD-10 desalting columns) or desalting spin columns (such as Zeba Spin desalting columns) to remove salt from the sample. These methods achieve desalting by gel filtration, separating large protein molecules from small salt ions.


2. Precipitation Method:

Use alcohol precipitation, chloroform-methanol precipitation, and similar methods to precipitate the protein, then resuspend it in deionized water or a low-salt concentration buffer. This can reduce the salt content in the sample to some extent.


3. Reverse Phase Chromatography:

Use high-performance liquid chromatography (HPLC) for protein separation and purification. Reverse phase chromatography can effectively remove salt from samples and purify proteins. This method is suitable for samples requiring high selectivity and resolution.


4. Ion Exchange Chromatography:

Ion exchange chromatography can separate proteins and salts by exploiting the electrostatic interaction between proteins and ion exchange resins. This method can reduce the salt content in samples and purify proteins.


5. ZipTip Method:

During pre-mass spectrometry processing, the ZipTip method can be used for sample desalting. The ZipTip is a convenient and fast method for processing small-volume samples, using a miniature reverse phase chromatography column for desalting and concentration.


When dealing with samples containing salt, the appropriate desalting method can be selected based on the sample characteristics, experimental conditions, and experimental requirements. Removing salt from samples helps improve the accuracy and reliability of mass spectrometry analysis.
