List the main methods for separating and purifying proteins and briefly explain their principles

The following are some common methods for protein separation and purification along with their brief principles:


1. Salting Out:

By gradually increasing the salt concentration (such as ammonium chloride), proteins precipitate from the solution. Different proteins have varying solubility with changes in salt concentration, allowing for partial purification.


2. Electrophoresis:

Separation is achieved based on protein migration behavior in an electric field. Common electrophoresis methods include one-dimensional or two-dimensional polyacrylamide gel electrophoresis (SDS-PAGE) and isoelectric focusing (IEF).


3. Liquid Chromatography:

Separation is based on differences in interaction between proteins and chromatographic column materials. Common liquid chromatography methods include:


1. Affinity Chromatography:

Separation is based on highly specific interactions between proteins and specific ligands.


2. Ion Exchange Chromatography:

Separation is based on interactions between protein surface charges and ion exchange resins.


3.Size Exclusion Chromatography (SEC):

Separation is based on protein size distribution within gel particles.


4. Hydrophobic Interaction Chromatography (HIC):

Separation is based on hydrophobic interactions between proteins and hydrophobic resins.


4. Ultrafiltration:

Uses a semi-permeable membrane to separate proteins with different molecular weight cut-offs (MWCO). Large proteins are retained on one side of the membrane, while small molecular impurities pass through into the filtrate.


5. Precipitation Method:

Exploiting protein instability under specific conditions (such as low temperature, high concentrations of alcohol, or changes in pH) to precipitate them. The precipitated proteins can be separated by centrifugation.


These methods can be used individually or in combination to achieve higher purity proteins. The choice of method depends on the properties of the target protein, its source, and the experimental purpose.
