Single-Cell Mass Cytometry Analysis: How to Measure Volume and Size, What is the Principle?

Single-cell mass cytometry (CyTOF) combines flow cytometry and mass spectrometry technology. It is mainly used to analyze molecules on the cell surface and within cells. Flow cytometry can measure the volume and size of cells by detecting individual cells passing through the flow channel using lasers or other light sources. The measurement of cell volume and size is mainly based on the following principles:

1.Forward Scatter (FSC):

FSC is related to the size of the cell. When a laser beam in the flow cytometer hits a cell, the cell scatters some of the laser light forward. Larger cells scatter more light, so FSC is typically used to estimate the size of cells.

2.Side Scatter (SSC):

SSC is related to the complexity or granularity of the cell. After the laser beam hits the cell, particles within the cell scatter light to the side. SSC can be used to estimate the granularity or internal complexity of the cell structure.

3.Dye Measurement:

Some flow cytometry techniques can measure cell volume using dyes. These dyes selectively bind to the cytoplasm, and the amount of binding is proportional to the cell volume. By measuring fluorescence intensity, the cell volume can be estimated.
