How is protein extraction done? The sample is culture medium with exogenously added proteins.
1. Sample Collection and Preparation:
First, collect samples from the culture medium. Ensure that the samples contain the desired exogenous protein.
Remove cell debris and unused culture medium using centrifugation and other methods.
2. Protein Lysis:
Use a lysis buffer (usually containing certain detergents and salts) to disrupt cell membranes, releasing proteins into the solution.
Sometimes, enzymes (such as trypsin) or physical methods (such as ultrasound) may be needed to assist lysis.
3. Protein Extraction:
Centrifuge the lysed mixture to separate the protein-containing supernatant.
Proteins can be further purified using methods such as salting out, dialysis, and affinity chromatography.
4.Protein Quantification and Analysis:
Use methods such as the Bradford, Lowry, or BCA protein assay to quantify the extracted proteins.
Techniques such as SDS-PAGE gel electrophoresis and mass spectrometry are used for protein analysis and identification.
