What is DTNB dissolved in? Is the solution neutral or alkaline during the thiol testing process?
DTNB (5,5'-dithiobis(2-nitrobenzoic acid), also known as Ellman's reagent) is typically dissolved in ethanol or water. During the assay of thiols (sulfhydryls), the reaction with DTNB usually occurs in a near-neutral or slightly alkaline environment. This is because under such pH conditions, thiol groups can react more effectively with DTNB.
