Today when doing WB, I first boiled the sample, then added the loading buffer, and put it in the refrigerator. Is it okay to boil it again when I need to use it?
Yes, if the sample has been boiled once and stored in the refrigerator, it is recommended to reheat it before use to ensure that the components in the loading buffer, such as SDS and DTT (or β-mercaptoethanol), can fully function to completely denature the proteins. Before reheating the sample, ensure it is fully thawed and gently mixed to avoid layering or precipitation of the components, which could affect the experimental results.
