Quantitative ubiquitination proteome analysis
Quantitative ubiquitination proteome analysis involves the quantitative analysis of ubiquitinated proteins at the omics level. Ubiquitin is a highly conserved polypeptide composed of 76 amino acid residues, which can form covalent bonds with one or more lysine residues on target proteins within cells under the action of enzymes. Ubiquitinated proteins are proteins linked with ubiquitin. Protein ubiquitination is involved in various life activities and is also related to the pathogenesis of diseases. Quantitative analysis of ubiquitinated proteomes using mass spectrometry
Mass spectrometry can be used to achieve quantitative analysis of ubiquitinated proteomes. Due to the low abundance and wide dynamic range of ubiquitinated proteins in biological samples, enrichment of ubiquitinated proteins and peptides is typically required before mass spectrometry analysis to enhance their abundance. Traditional quantitative proteomics methods are then used to perform quantitative analysis on the enriched ubiquitinated peptide samples. In mass spectrometry-based ubiquitination quantitative proteomics research, the ubiquitinated proteins are first digested with trypsin, generating two glycine residues at the modification sites on lysine. Specific enrichment of ubiquitinated peptides is performed using a kit targeting these residues. Subsequently, samples are pre-separated using HPLC, and ubiquitinated peptides are enriched using antibodies against special residue structures, greatly improving the identification of ubiquitinated proteins and thereby enhancing the quantification of the ubiquitinated proteome.
