Label-free proteomics
Based on whether labeling techniques are used, mass spectrometry-based proteomics research can be divided into label-free and labeled methods. Label-free proteomics uses LC-MS to analyze proteolytic peptides without the need for expensive stable isotope labels as internal standards. By analyzing the mass spectrometry data generated from large-scale protein identification, comparing the signal intensities of corresponding peptides in different samples allows for relative quantification of the proteins corresponding to the peptides.

Label-free techniques are commonly used for quantitative proteomics analysis. Label-free quantitative proteomics analysis not only quantifies the proteome but also qualifies it. Compared to labeled quantitative proteomics, label-free quantitative proteomics is more cost-effective, suitable for analyzing relatively simple protein mixtures, and is also ideal for large sample quantitative comparisons and samples that cannot be analyzed using labeled quantification.
