Choosing MALDI-TOF/TOF or LC-MS/MS for Proteomics?
If the band is Coomassie-stained, the fastest and most cost-effective method for protein identification is mass spectrometry (MS/MS sequencing), choosing MALDI-TOF/TOF (PMF+MS/MS) mass spectrometry and automatic database analysis. If the band is silver-stained or may contain more than one protein, the highest sensitivity is achieved using mass spectrometry (via MS/MS sequencing) and electrospray (LC-MS/MS) mass spectrometry for protein identification, accompanied by automatic database analysis.
