What are the different types of immunoprecipitation?

Single Protein Immunoprecipitation: This method involves isolating one protein from a solution containing several other proteins. The sample range includes crude extracts of animal and plant cells, and body fluids. In single protein immunoprecipitation, the target protein is known, and specific antibodies are used to isolate the protein. Protein Complex Immunoprecipitation: This method involves the immunoprecipitation of complexes containing the antigen and related proteins or ligands that bind to it. Protein complex immunoprecipitation is also known as co-immunoprecipitation. When several proteins are present in a complex and one of its members is known, unknown proteins can be identified with the help of antibodies that can bind to the known member. When the members of a protein complex are connected to each other, it is easier to isolate the entire complex and identify unknown members. This method is sometimes also referred to as a pull-down. Chromatin Immunoprecipitation: Chromatin Immunoprecipitation, or ChIP, is a method for identifying protein-DNA interactions within cells. This method is used to understand which genomic regions are associated with specific proteins or whether several proteins are associated with a specific genomic region. ChIP assays can also be used to identify the order of protein recruitment in promoter regions or histone modifications of specific genomic regions. Additionally, ChIP provides information related to transcription factor binding. RNP Immunoprecipitation: Gene expression regulated by ribonucleoprotein complexes is post-transcriptionally controlled. Ribonucleoprotein complexes include RNA-binding proteins and non-coding RNAs, which together regulate cellular functions such as splicing, mRNA stability, export, or protein localization. Similarly, mRNAs involved in protein synthesis for specific cellular processes are found to be associated with ribonucleoprotein complexes forming mRNPs. These RNA-binding proteins, which are crucial for RNA functions, are also important targets for potential therapeutic interventions. Therefore, with the help of antibodies against RNA-binding proteins, RNP Immunoprecipitation or RNP-IP can be used to isolate miRNA and mRNA from ribonucleoprotein complexes.
