What is the difference between GST-pull down and Co-immunoprecipitation?
Immunoprecipitation is used for precipitating proteins in their natural state, maintaining the complex state of proteins as they represent their natural form. However, Co-IP cannot exclude whether the proteins present in the complex interact directly or through a third protein. In contrast, the GST pull-down assay is an in vitro method for examining protein-protein interactions. In the GST-pull down analysis, a glutathione S-transferase tag is added to the N-terminus of the target gene, which produces the desired protein. Next, this protein is pulled down by glutathione beads due to their high affinity for GST. If the protein of interest is part of a complex with other proteins, these other proteins will also be pulled down with the target protein.
