How to choose the membrane for western blotting?
When selecting a membrane, factors such as the type of membrane and pore size should be considered. Compared to nitrocellulose membranes (80-100mg/cm2), PVDF membranes have a higher protein binding capacity (170-200mg/cm2), making them more suitable for detecting low-abundance proteins, although they may lead to higher background noise. The typical pore sizes for both PVDF and nitrocellulose membranes are 0.1, 0.2, or 0.45mm, with 0.45mm being commonly used. Nitrocellulose membranes are ideal for detecting low molecular weight proteins, while PVDF is better suited for high molecular weight proteins. For fluorescent multiplex immunoblotting, low-fluorescence PVDF membranes should be used to reduce background levels under blue and green light.
