What is the void volume in size exclusion chromatography (SEC)?
In SEC (Size Exclusion Chromatography), the void volume is the volume of mobile phase required for an eluting molecule to have a zero retention rate in the stationary phase. Ideally, it equals the volume of the mobile phase in the column, which is the volume of the pores and spaces between the stationary phase. The void volume is usually expressed as a percentage of the total column volume. For example, if the stationary phase particles occupy 70% of the total column, the void volume will account for 30% of the total column volume.
