What is the function of SDS in SDS-PAGE?
SDS (Sodium dodecyl sulfate) is an anionic detergent that can unfold and denature proteins and wraps proteins in a negative charge. SDS is added in excess to proteins to cover their intrinsic charges so that all proteins acquire a similar charge-to-mass ratio. In this way, the migration rate of proteins will depend on their size rather than their intrinsic charge.
