Ion Exchange Chromatography - How to Choose Strong/Weak Ion Exchange Resins for Protein Purification?
First, choose a strong ion exchanger. If the selectivity is not good enough, try using a weak ion exchanger. Strong ion exchangers (Q, SP, and S) are more stable over a wider pH range, while the stability of weak ion exchangers (DEAE, ANX, and CM) varies with pH. Both cation and anion exchangers can be used for protein purification, and selecting one may be more effective for a specific protein. If the protein's isoelectric point (pI) > 7, a cation exchanger is suitable; if pI < 7, an anion exchanger can be selected.
