Co-Immunoprecipitation

Co-Immunoprecipitation (Co-IP) is an analytical technique developed from immunoprecipitation to study protein interactions, serving as a supplement and extension of the immunoprecipitation technique. Many beginners might not be very familiar with Co-IP, so today I'll guide you through understanding the technical principles of Co-Immunoprecipitation.

Co-Immunoprecipitation is based on the principle of specific binding between antibodies and antigens. It uses specific antibodies to specifically recognize one of the interacting proteins in a mixed system, and then precipitates them together to identify the interaction between two known proteins or the interaction between a known protein and an unknown protein. This technique uses non-denaturing agents to lyse cells, preserving the interaction between proteins A and B within the cell. Then it is mixed with specific antibodies for protein A or B, where the antibody binds to protein A or B to precipitate it, while protein B or A is co-precipitated. This technique of immunologically co-precipitating two interacting proteins in one go is called Co-Immunoprecipitation.
