Host Cell Protein (HCP) Detection Methods and Principles
HCP (Host Cell Proteins) refers to protein impurities from the cells used in the production of biopharmaceuticals. For example, when using mammalian cells like CHO cells to produce recombinant proteins, besides the target protein, there may also be proteins from the CHO cells present in the final product. These HCPs can affect the stability, efficacy, or safety of the product, thus requiring detection and control.
Below are common methods for HCP detection and their principles:
1. ELISA (Enzyme-Linked Immunosorbent Assay)
(1) Principle: Specific anti-HCP antibodies are immobilized on an assay plate. HCPs in the sample bind to these antibodies, and then enzyme-labeled secondary antibodies that bind to HCPs are added. By measuring the enzyme activity, the HCP content can be indirectly measured.
(2) Advantages: Sensitive and capable of quantifying the total amount of HCPs.
(3) Disadvantages: Requires high-quality anti-HCP antibodies and may not detect all HCP impurities.

2. Mass Spectrometry
(1) Principle: Using a mass spectrometer to detect and identify proteins in the sample.
(2) Advantages: Can identify specific types of HCPs, high resolution.
(3) Disadvantages: Technically demanding, high cost, complex data processing.
3. Immunoblot (Western Blot)
(1) Principle: Proteins are first separated by gel electrophoresis, then transferred onto a membrane, and specific anti-HCP antibodies are used to detect target HCPs.
(2) Advantages: Can identify and quantify specific HCPs.
(3) Disadvantages: Only detects HCPs that react with the antibodies used.
4. Two-Dimensional Gel Electrophoresis (2D-PAGE)
(1) Principle: Proteins are separated through two-dimensional gel electrophoresis, first by isoelectric point and then by molecular weight, creating a two-dimensional protein map.
(2) Advantages: Can observe the distribution of all proteins in the sample.
(3) Disadvantages: Technically demanding, cannot directly quantify.
During the development and production of biopharmaceuticals, multiple methods are usually combined to detect and control HCPs to ensure product quality and safety.
