I. Principle
The fundamental principle of hydrogen/deuterium exchange mass spectrometry is that hydrogen atoms in a protein can exchange with deuterium atoms in the solution. This process is influenced by various factors, including the pH and temperature of the solution, as well as the conformation of the protein. By measuring the rate of this exchange process, we can understand the protein conformation under these conditions.
II. Methodology
In a typical hydrogen/deuterium exchange experiment, the protein is first incubated in a deuterium-containing solution for a period of time, allowing the hydrogen atoms to exchange with deuterium atoms. The protein is then rapidly frozen and analyzed by mass spectrometry to measure the deuterium content on each amino acid residue.
By comparing the deuterium content at different time points, we can calculate the hydrogen/deuterium exchange rate for each amino acid residue, thus obtaining information about protein conformation. For example, if the exchange rate of an amino acid residue is slow, it may be located inside the protein or form hydrogen bonds with other amino acids; if the exchange rate is fast, it may be located on the surface of the protein.
III. Applications
Hydrogen/deuterium exchange mass spectrometry has applications in many fields. For instance, it can be used to study protein folding and unfolding processes, analyze interactions between proteins, and investigate drug-protein binding.
Combined with other techniques, such as electron microscopy and NMR, hydrogen/deuterium exchange mass spectrometry can provide additional information about protein conformation.
