How to perform sample pretreatment for MALDI TOF mass spectrometry analysis?
Before performing MALDI-TOF MS analysis, sample pretreatment is an important step as it directly affects the quality and accuracy of the analysis. Common sample pretreatment steps include:
1. Sample purification:
Samples need to be highly purified to reduce interference from impurities, as they may inhibit the ionization process or interfere with mass spectrometry signals. Impurities can be removed through centrifugation, gel filtration, or dialysis.
2. Sample concentration:
If the sample concentration is too low, methods such as using concentration centrifuge tubes or evaporation may be required to increase detection sensitivity.
3. Protein or peptide digestion (if needed):
For protein samples, enzymatic digestion (e.g., using trypsin) is usually needed to produce peptides for analysis. The digestion process requires precise control of conditions such as temperature and time.
4. Matrix selection and preparation:
Selecting the appropriate matrix is a key step because the matrix absorbs laser energy and transfers it to the sample. Common matrices include α-cyano-4-hydroxycinnamic acid (CHCA), octanoic acid, and dihydroxybenzoic acid. Matrix solutions are typically prepared in organic solvents such as ethyl acetate, methanol, or ethanol.
5. Mixing the sample with the matrix:
The sample is mixed with the matrix solution, usually at a certain ratio (e.g., 1:1). After mixing, the solution is spotted onto the MALDI plate to dry naturally or with gentle heating.
Each of the above steps requires precise control to ensure the representativeness of the sample and the accuracy of the analysis results. Additionally, the specific steps of sample processing may vary depending on the sample type and analysis purpose.
