Introduction to Proteomics MRM
MRM (Multiple Reaction Monitoring) is a targeted quantitative proteomics technology used to study target protein molecules. MRM is based on known or hypothesized information about target molecules, allowing for the selective choice of ions for mass spectrometry data acquisition. It collects signals for ions that meet the target ion criteria while eliminating interference from non-compliant ion signals. MRM mass spectrometry analysis includes three stages: 1. Selecting precursor ions specific to the target molecule using MS; 2. Fragmenting these precursor ions to remove interference from other ions; 3. Collecting mass spectrometry signals only for the selected specific MS/MS2 ions. MRM mass spectrometry technology is a highly accurate protein quantification and identification method, making it an excellent approach for the precise quantification of multiple target proteins in complex samples. By using isotope-labeled target peptides as internal standards, it is possible to achieve absolute quantification of proteins.

MRM technology, through two levels of ion selection, can exclude a large number of interfering ions, thereby lowering the background of the mass spectrometry and significantly improving the signal-to-noise ratio of the target analyte. This results in high sensitivity detection with good reproducibility and high accuracy. It is particularly suitable for detecting differences in protein expression levels of known protein sequences and can detect proteins with relatively low abundance. The difference between MRM and PRM is that after fragmenting the selected precursor ions, MRM measures only the selected fragment ions, while PRM measures all fragment ions.
